| Research Abstract |
哺乳動物の副腎皮質は、球状層、束状層、網状層の3層からなることが従来より知られていた。我々は、3層に加えて、球状層と束状層の間に未分化な細胞層(以下、未分化細胞層)を近年新たに見い出した。この細胞層の種々の性質から、未分化細胞層は、多能性の幹細胞を含み、それが分化・増殖して球状層と束状層の細胞が生じるとの仮説を提唱した。本研究課題は、この仮説を証明するため、副腎皮質から得られた単一クローンから成る未分化な細胞集団から、分化した副腎皮質細胞が生じることを示す。
本年度は、温度感受性SV40T抗原遺伝子をトランスジーンとして持つマウス副腎から、副腎皮質の性質を持つ細胞株の樹立を進めた。このマウスは、正常に発生し、その副腎は未分化細胞層を含めて、形態的機能的に正常マウスとの違いは認められない。このことから、組織の細胞は、正常な発生・分化の途上にあると考えられる。許容温度で樹立した細胞株について、副腎皮質の細胞層マーカーの発現を検出したところ、大部分が束状層様の性質を保持していた。一部の株は、皮質の全細胞に存在するマーカーを発現するが、層特異的マーカーを発現しないことから、未分化細胞層と同様な性質を保持していることが判明した。異なる形質の細胞株が得られたことから、細胞株の性質は、組織から取り出した時点の分化形質を反映していることが期待された。未分化細胞株の性質を詳細に解析した結果、非許容温度でdibutyryl cAMPの存在下で培養することにより、束状層マーカー遺伝子の発現が誘導されることが判明した。つまり、未分化な状態から、束状層様に変化することが判明した。この変化は不可逆的であることから、分化と考えられる。このことから、少なくとも未分化細胞層様の細胞株が、束状層細胞様に分化しうることが明らかになった。
