| Research Abstract |
平成12年度の研究計画に基づいて下記の研究結果を得た。
1.多発性骨髄腫においてノンランダムに認められる染色体転座であるt(14;20)(q32;q11)の20q11領域の転座切断点がFISH法によりKLAA0823とMAFB遺伝子の間の680kb以内に存在することを明らかにした。同領域のPACクローンを用いてエクソントラップ法等を用いて標的遺伝子の同定を試みた結果、免疫グロブリン遺伝子のαエンハンサーによる脱制御を受けてMAFB遺伝子が異所性に発現していることを見い出した。同時にt(14;20)転座を検出する二重色FISH診断法も確立し、臨床検体での解析を開始している。
2.1と同様にt(1;14)(p36;q32)転座の1p36領域の切断点がE3/LAPTM5遺伝子内に存在することを見い出した。現在、転座による遺伝子発現抑制の機序および切断点近傍に別の標的遺伝子が存在するか否かについても検討を進めている。
3.骨髄腫の前癌病変であるMGUSおよびくすぶり型骨髄腫症例20例の骨髄液からインフォームドコンセントを得た後、抗CD138抗体ビーズを用いて形質細胞を単離した。90%以上の純度を有する試料につきカルノア検体を作成した。4.N/K-RAS遺伝子の変異を容易にスクリーニングするPCR-RFLP法を確立した。また、JCOG-LSGグループにおいて登録される骨髄腫症例において腫瘍細胞に認められるこれらの体細胞遺伝子変異解析を行うための研究計画書の作成を行っている。
5.骨髄腫細胞株15株において染色体不安定性に関与することが報告されているKu80遺伝子の変異の有無、遺伝子再構成の有無およびmRNA、蛋白レベルでの発現を調べた。2アレルで異常を認めた細胞株は認めず骨髄腫発症への関与は少ないものと結論した。
