Research Abstract

新しい成長ホルモン分泌促進ペプチド、グレリンに関して、本年度中に申請者は次のような研究を行った。
1,オーファン受容体GHS-Rの内因性リガンド、グレリンを胃から精製し、構造決定した。グレリンがin vitro,in vivoで成長ホルモン分泌を刺激することを明らかにした。ヒトにおいても、グレリンの静脈内投与によっても強力な成長ホルモン分泌活性をもつこと明らかにした。2,グレリンの第2の内因性formとしてdes-Gln14-ghrelinを精製・構造決定した。des-Gln14-ghrelinはグレリンと同一の遺伝子から選択的スプライシングによって、生成されることを明らかにした。des-Gln14-ghrelinはグレリンと同じく、血中投与によって強力な成長ホルモン分泌作用を示した。3,グレリンの胃・消化管での分布を免疫組織染色で調べた。その結果、30年以上不明であった胃のX/A-like細胞の分泌顆粒の中身が、グレリンであることを明らかにした。4,視床下部の摂食中枢を介して、グレリンが摂食活動を促し、体重増加をもたらすことを明らかにした。5,グレリンが胃において胃酸分泌、胃液分泌亢進作用を示すことを明らかにした。6,グレリンのRIA測定系を確立した。
