Research Abstract	本研究の目的は軟骨肉腫に対して遺伝子治療を実験的に行い臨床応用への基盤を作ることにある。平成12年度よりの研究でレトロウイルスベクターを用いて単純ヘルペスウイルスのチロシンキナーゼの軟骨肉腫細胞への遺伝子導入と抗ウイルス薬の投与による殺細胞効果を細胞培養系で確認した。さらに、遺伝子導入細胞の死は非導入細胞を巻き添えにすることも確認してきた。本年度の研究では巻き添え殺細胞効果は1個の遺伝子導入軟骨肉腫細胞が周囲に存在する約20個の非遺伝子導入細胞を死に至らしめることを示した。さらに、軟骨肉腫担がんヌードマウスに対して、腫瘍内に遺伝子導入軟骨肉腫細胞を注入し抗ウイルス薬を投与することにより、in vivoで腫瘍の縮小効果がみられることを示した。ヌードマウスの背部腫瘍が約250mm^3になったところで10^6個の遺伝子導入軟骨細胞を注入し、その後抗ウイルス薬を週2回2週間投与することにより腫瘍の大きさは非投与群の4分の1まで抑制されることを明らかにした。組織学的には腫瘍の広範な壊死像を認めた。また、X-gal染色で注入された遺伝子導入軟骨肉腫細胞が腫瘍組織内に存在していることも確認し、巻き添え殺細胞効果(bystander effect)が動物の生体内でも存在することを示した。3年間の研究期間での成果は軟骨肉腫の遺伝子治療は臨床でも有効である可能性を示唆したものである。骨盤や体幹発生の外科的切除の不能な軟骨肉腫では遺伝子治療を導入することができるものと考えられた。
