| Research Abstract |
<研究の目的>
我々は日本人男性のY染色体を4つに分類し,タイプによってヒト精子数が有意に異なること,また精子数の少ないタイプは他のタイプに比べて無精子症になりやすいことを報告した.この研究の過程でY染色体のタイプによって精子数以外の表現型が違うものが存在するか,また実際の歴史の中で,それぞれのタイプの男性の割合がどのように変化してきたかに関心を抱いた.これらについて,ヒトの個体差,ヒト集団の変化という点を分子生物学的知見に基づき明らかにすることが本研究の目的である.
<結果>
本年度は主として,迅速にY染色体ハプロタイプを決定するためのシステムの構築を行った.PCR, PCR-DHPLC (denaturing high performance liquid chromatography),自動シークエンサーを用いた蛍光SSCP(single strand polymorphism)を併用することで短時間に大量の検体を解析することができるようになった.また実際に多数の男性から微量血液検体を採取した場合を想定して,微量血液からゲノムDNAを抽出し,固相化することでPCR反応後にゲノムDNAが再利用できるような技術の検討も行った.我々の検討では少なくとも3回のゲノムDNAの再利用が可能であった.今後,今年度構築したDNA多型解析のシステムを用いて多数のDNAサンプルのハプロタイピングを行い,Y染色体が男性表現型と適応度に及ぼす影響を明らかにする予定である.
