Research Abstract

本研究では、最近相次いで発見されているペプチド・ビルトイン型のキノン補酵素を含有する酵素の構造と機能を明らかにするとともに、タンパク質翻訳後修飾によるキノン型ビルトイン補酵素の生合成機構を解明することを目的としている。グラム陰性細菌のPseudomonas putida及びParacoccusdenitrificansは、培地中にn-ブチルアミンやベンジルアミンを加えると、これらをエネルギー源として資化するため、キノヘムプロテイン・アミン脱水素酵素(QHAmDH)を細胞膜内のペリプラズム画分に誘導生成する。本酵素は、αβγのヘテロ三量体サブユニット構造をもち、約60kDaのα-サブユニットには2分子のヘムcが、約9kDaのγ-サブユニットには構造未知のビルトイン型キノン補酵素が含まれている。触媒反応においては、基質アミンに由来する2電子はキノン補酵素、ヘムcを経由して、アズリンやチトクロムc550などの電子受容体タンパク質に受け渡され、最終的には末端酸化酵素により分子状酸素の水への還元に使われる。平成13年度は、Ps.putida及びP.denitrificansのQHAmDHに含まれるキノン補酵素を同定するため、両細菌の酵素のX線結晶構造解析を行った。その結果、両細菌の酵素がともに新規なペプチド・ビルトイン型キノン補酵素、システイントリプトフィルキノン(CTQ)を含有することを解明するとともに、CTQを含む小サブユニットが4ヶ所の分子内チオエーテル結合を含む極めて特異な構造を有することを明らかにした。
