高圧温度ジャンプ法と計算機シミュレーションによるタンパク質フォールディング研究

Research Abstract

本研究計画では、蛋白質フォールディング機構解明を目的として、以下の研究を行う。
(1)高圧条件下でのジュール熱方式温度ジャンプ法を用いて、スタフィロコッカル・ヌクレアーゼ(SNase)、α-ラクトアルブミン(αLA)及びカルシウム結合性リゾチームの巻き戻り速度過程をマイクロ秒からミリ秒の時間域で追跡する。(2)αLA、カルシウム結合性リゾチーム及びSNaseを対象に、フォールディング過程の原子レベルでの記述を行う。そのため、分子動力学法による高温下(400〜600K)でのアンフォールディング・シミュレーションを行う。
本年度の研究により、2000気圧の高圧下で作動可能なジュール熱方式温度ジャンプ装置をユニソク社とともに設計製作した。トリス緩衝液-フェノールフタレインの温度変化に伴う光吸収変化を用い所期の温度ジャンプ幅が達成されていることを確認した。また、圧力も2000気圧の高圧に十分耐え得ることを確認した。高圧条件にすることの目的は、水の氷点を-20℃まで下げ、低温変性条件下での高速温度上昇に伴う蛋白質の巻き戻り反応を調べることにある。また、巻き戻り測定には、光吸収のみならず芳香族側鎖の蛍光スペクトルも用いる。製作した装置は、これらの点においてまだ性能が十分ではなかったので、現在改良加工を加えている。
野生型ヤギαLAの600K下でのアンフォールディングの分子動力学シミュレーションを行った。150から200ピコ秒のシミュレーションでアンフォールディングの初期過程に至ることを確認した。今後、アンフォールディング速度を著しく変えることが実験的に確認されている変異タンパク質についてアンフォールディングシミュレーションを行い、実験結果との対応関係を明らかにする。
