Research Abstract

多くの細菌は、低栄養、低温などのストレス条件下におかれると、異なる生理状態に移行する。そうした状態のひとつとして、培養できないが、生理活性を維持した状態がある。本研究は、培養不能状態にある細菌の生理状態を、分子レベルで解明することを目的としている。その研究実績のいくつかを述べる。
第一に、熱水や冷湧水が供給される深海底また温泉などの化学合成生態系は地球上での細菌の進化を考慮する上で鍵となる生態系である。これらの環境中の細菌群集の培養能を調べるとともに、その種組成を培養によらない方法を適用して調べた。第二に、海洋には細菌の10-100倍のウイルス様粒子が存在し、海洋細菌に感染することで、その死滅や培養能の喪失を招く。これらのウイルス様粒子を天然海水から濃縮、分離し、その機能、遺伝子伝播に与える影響について基礎的知見を得た。第三に、培養不能状態への移行あるいはそこからの復帰には活性酸素によるストレスが大きく関与していると予想されてきた。培養不能状態の菌に対してこうしたストレス状態が異なる条件を与え、実際にそれが培養能を左右していることを明らかにした。第四に、培養不能状態の菌は培養可能な菌や死滅した菌と共存していることにより、その解析が困難とされてきた。Percoll密度勾配法により、異なる培養能、遺伝子発現の状況下にある集団を分離、解析する新たな手法を開発した。第五に、大腸菌が対数増殖期から定常期に移行する際に、どのようなシグマファクターがどのような遺伝子群の転写に関与しているかについての基礎知見を得た。
