Research Abstract

内分泌かく乱化学物質は低用量での用量-作用関係が未だ明らかにされておらず、暴露量の把握を含めて正確なリスク評価を導き出せない状況にある。精度の高いリスク評価を実施するためには、人体暴露量評価が重要であり、このためには血液等の生体試料の分析や様々な暴露媒体を分析して存在量を推定する必要がある。本年度は、生活関連製品に由来し、様々な媒体を介して体内に取り込まれる可能性が危惧されている内分泌かく乱物質を取り上げ、リスク評価に資するデータを取得することを目的とした。
(1)内分泌かく乱化学物質の生体試料の分析と体内動態の解明(中澤・加藤・井之上・吉村)
フタル酸エステル類、ビスフェノールA(BPA)等について血液、尿等の生体試料中の当該化学物質及びその代謝物の分析法をLC/MS、LC/MS/MS等を用いて高精度かつ高感度な分析法を構築し、体内における動態を解明した。
(2)生活環境並びに生活関連製品中におけるフタル酸エステル類、BPA及び関連化合物の動態解明(井之上・吉村・加藤・中澤)河川水、飲料水中に含有されるBPAを含むフェノール製化合物を測定した。また、食品用容器包装資材等から食品に移行するフタル酸エステル類、BPA等の分析法をLC/MS等を用いて構築した。開発した分析法を用いて蜂蜜等の食品中のBPAおよび関連物質を測定した。更に食品に移行したフタル酸ジ2エチルヘキシル(DEHP)の食品中における分解挙動や塩化ビニル製医療用具から溶出され、血液や輸液等への移行が危惧される。DEHPについて分析法を開発し、実試料を分析して、動態を明らかにした。また、当該化学物質の動物実験を用いた生体影響の評価データには飼育環境からの汚染が懸念されており、フタル酸エステル類のバックグランドレベルを把握した。
(3)内分泌かく乱作用の評価について(井之上・吉村・加藤・中澤)
内分泌かく乱作用の評価にエストロゲン受容体結合試験、乳がん細胞増殖試験を駆使して、食品用塩化ビニル製ラップフィルム中の残留化学物質やBPA関連化合物のエストロゲン活性を評価した。
