| Research Abstract |
我々は、アジア人種に高率に認められる視神経脊髄型多発性硬化症(opticospinal multiple sclerosis,OS-MS)の疾患感受性遺伝子がHLA-DPB1^*0501遺伝子であることを発見した。さらに主要なミエリン抗原(MBP,PLP,MOG)に反応性を示すT細胞株を樹立した場合、OS-MSと通常型MSとでは、エピトープの拡大する傾向が抗原蛋白により異なることを見いだした。このミエリン抗原に対する反応性の違いは、MSの2病型のそれぞれの疾患感受性を示すHLAクラスII遺伝子に由来するものと考えられた。また、近年オリゴデンドログリアの起源は単一でなく、視神経・脊髄と大脳ではlineageが異なる可能性が指摘されており、OS-MSの病巣の形成には、視神経・脊髄に特異的な自己抗原が存在する可能性が考えられた。これらのことより以下のことを行った。ヒト脊髄Poly A+RNA(Clontech)を購入し、ランダムプライマーを用いてcDNAを合成、末端を平滑化した後、アダプターをライゲーションしλZAP II Vectorにベクターライゲーションを行った。これによりヒト脊髄cDNA発現ライブラリーが作製できた。この発現ライブラリーを用いて、SEREX法にて自己抗原のスクリーニングを開始した。OS-MS患者1名の血清より、およそ5×10^5個のファージのスクリーニングにより7つのポジティブクローンが検出され、クローニング後インサートの配列を決定した結果、7つのうち3つはframe shiftしたネガティブクローンであったが、2つはRabaptin-5蛋白、残り2つはその機能が不明のKIAA0610蛋白であった。現在、この2つの蛋白のOS-MSに対する意義の検討と、さらに他のOS-MS患者でのSEREX法によるスクリーニングを進めている。
