悪性骨軟部腫瘍の浸潤、転移に関わるシグナル伝達分子を標的とした新しい治療法の開発

Research Abstract	悪性骨軟部腫瘍患者の死因のほとんどは転移である。転移に際し腫瘍細胞は血管内皮細胞下の基底膜を浸潤、破壊するが、この基底膜浸潤は、転移成立の多段階プロセスのなかで最も重要と考えられている。我々は、この基底膜浸潤のモデルであるinvasion assayを開発し、基底膜浸潤の分子機構を解析し、その抑制法の開発に向けた研究を推進してきた。その結果、悪性腫瘍細胞の基底膜への接着、酵素的破壊、運動に関与する遺伝子は、接着分子や細胞骨格関連分子からのシグナル伝達により発現が一挙に変化し、浸潤、転移を惹起している可能性が示唆された。一方、近年、Rhoファミリー、インテグリンという浸潤、転移に関与する2つの主要な分子が注目され、その下流のシグナル伝達経路の解析が進んでいるが、両者の相互関係は全く明らかにされていない。今回の研究の目的は、インテグリンとRhoのシグナル伝達経路とクロストークの詳細な検討を行い、その経路の中で細胞の転移形質を一度に抑制しうる最も重要な因子を分子標的とし同定し、その分子標的に対する有効な抑制法を検討し、悪性骨軟部腫瘍の浸潤、転移の新しい治療法の開発につなげることである。本年度は、骨肉腫細胞株にRho活性化剤を作用させ、過剰なRho活性化は細胞運動、浸潤能の低下、基底膜破壊に重要な酵素であるMMP2の活性化抑制を一挙に起こすことを証明した。このRho活性化は細胞接着分子インテグリンの下流で、細胞運動、浸潤能を制御しているFAKのリン酸化も抑制した。また、転移性の悪性組織性組織球腫の臨床サンプルにおいてもRhoが過剰発現していることを見いだした。さらに、骨転移成立過程において、血管内皮細胞増殖因子が腫瘍の血管新生促進とともに、破骨細胞前駆細胞の増殖、運動能を亢進させること、その下流のシグナル伝達にFAKが関与していることを見いだした。
