Research Abstract	標的遺伝子のmRNAに相補的なオリゴヌクレオチド(ODN)を細胞へ投与し、当該遺伝子の発現のみを特異的に抑制するアンチセンス法は新しい疾病の治療法として注目を集めている。本研究では、糖部立体配座を固定した新規ヌクレオシド類縁体(BNA)をODNへ導入し、アンチセンス分子として必要とされる種々の特性について詳細な評価を行うと共に、新たなBNA類の開発を推進し、以下のような成果を得た。 1. BNA修飾型アンチセンスODN機能評価標的として設定したがん関連遺伝子、ウイルス遺伝子に対するアンチセンスODN中へBNAを導入し、その化学的・生物学的特性について評価した。まず相補鎖RNAに対する結合親和性を核酸融解温度(Tm)測定により検討したところ、いずれの場合もBNA導入により標的mRNAとの結合親和性は大きく向上した。さらに、これらBNA-ODN類が血清中で極めて安定であることを電気泳動により確認した。ODNへのBNA修飾の程度を調節することで、BNA-ODN/RNAヘテロ二重鎖がRNaseHより認識を受け、RNA鎖の分解を誘導することを明らかにすると共に、培養細胞系でのレポーター遺伝子の発現をBNA-ODNが極めて効果的に抑制することを見い出した。 2. N3'-P5'ホスホロアミダート結合を有する新規BNA-ODN類の開発 ODN中のリン酸ジエステル結合部をN3'-P5'ホスホロアミダート結合に置換することで、生体内での安定性等が大幅に向上することから、BNA型ODNへのN3'-P5'ホスホロアミダート結合の導入を行なった。既に合成に成功している3'-amino-2',4'-BNAモノマーを用い、1)DNA合成に適したヘテロダイマーを合成しODNへ導入する、2)通常とは逆の5'末端から3'末端への伸長反応を開発しODNへ導入する、という2種のアプローチを検討したところ、両手法とも目的の新規BNA-ODNを効率的に与えることを見い出した。
