旧ロシア核実験場セミパラチンスク周辺住民に多発する顎顔面奇形の分子疫学的研究

Research Abstract

カザフスタン共和国はユーラシア大陸の中央部に位置し,国境をロシア,キルギスタン,トルクメニスタン,中国およびカスピ海に接している。多くの科学者が警告したにもかかわらず,ソビエト連邦はカザフスタン共和国のセミパラチンスク地域で計458回にも及ぶ核実験を行った。1949年8月29日にセミパラチンスク市から約130km離れた核実験場(SNTS)での最初の核実験以来1989年までの40年間に地上30回,大気中88回および340回の地下核実験が行われた。その結果,セミパラチンスク周辺地域の数十万人にも及ぶ人々が様々な程度の放射線被爆を被った。
セミパラチンスク地域は14の経済区に分けられ,それら地区の核実験場からの距離は様々であり,放射線被爆の程度によって高線量被爆地域,中等度被爆地域および非被爆(コントロール)地域の3地域に分けることが出来る。高線量被爆地域の平均線量は1.0Sv(ジーベルト)以上,中等度被爆地域は0.5-1.0Sv,非被曝地域は0.03-0.07Svである。
本研究では,SNTS周辺の高線量被爆地域,中等度被爆地域およびコントロール地域の住民における唇顎口蓋裂の発症率の比較検討および,高線量被爆地域およびコントロール地域住民の口腔疾患および全身疾患罹患率の比較検討を行った。
その結果以下のことが明らかとなった。
1)高線量被爆地域,中等度被爆地域および非被爆(コントロール)地域における唇顎口蓋裂の発症率を統計学的に解析した結果,高線量被爆地域における唇顎口蓋裂の発症率は他の地域のそれと比較して有意に高いことが明らかとなり,被爆線量に比例して唇顎口蓋裂の発症率は上昇することが強く示唆された。
2)高線量被爆地域(サラジャール村)およびコントロール地域(コクペッティ村)における顎・顔面・口腔検診の結果を統計学的(ANOVA解析)に解析した結果,高線量被爆地域における喪失歯数,口腔疾患および全身疾患の発症率は非被爆(コントロール)地域と比較して有意に高いことが明らかとなった。
今後,今回の調査結果の生物学的背景を分子生物学的に検討し,放射線被爆と口腔疾患や先天疾患との関連性を明らかにしていきたい。
