Research Abstract

As to indole-3-acetic acid(IAA), endogenous levels by liquid chromatography-mass spectrometry, diffusible levels by fluorometric detection and polar transport activity by radioactive IAA were determined in 3-month-old tomato plants. IAA concentration in the apoplast(AP, sap outside a cell) was higher than those in the symplast (SP, sap within a cell) in both upper and lower parts of stems, showing that IAA analysis of AP is important. Younger leaves exported much more diffusible IAA than older leaves. IAA concentration in the main roots was almost at the same level as in the stems. It is suggested that at sampling, the transport capacity of IAA is probably the limiting factor for IAA transport in stems, and the amount of polar IAA transport might be only 19% of the endogenous IAA amount in stems.
The levels of IAA, gibberellic acid1 (GAl), trans-zeatin (Z) and trans-zeatin riboside(ZR) in seedless fruits of parthenocarpic tomato were analysed. Stage in 6cm diameter corresponding the middle of cell expansion phase were referred to expansion-stage, and stage in 8cm diameter corresponding the last of cell expansion phase were referred to MG-stage. IAA concentrations of the pericarps and partitions were higher in MG stage than in expansion-stage. In the partitions, IAA concentrations of SP were higher than those of AP in both expansion-and MG-stage. As to Z concentration, the SP of the partitions in expansion-stage had the highest and was 2.7 times than the AP, while in the pericarps were the same level in AP and SP. The ZR concentration in locule tissues in expansion-stage was the highest of all parts. The results suggest that the sites of synthesis may be the SP of partitions for IAA and Z, and locules for ZR.
