MPO遺伝子欠損を利用したMPO-ANCA関連血管炎の発症機構の解析

Research Abstract

本研究において、血管炎の発症に関連する好中球機能、myeloperoxidase(MPO)の性状、血清中の好中球抗体MPO-ANCA値について血管炎モデルマウスを用いて解析した。腎炎や血管炎発症モデルマウスは、自然発症のNZB/WF1、MRL、SCG、IRF-8/ICSBPマウスおよびCandida albicans-derived substances (CADS)誘発冠状動脈炎マウスを用いて解析した。また、血管炎にMPOおよびMPO-ANCAの関与は、われわれのグループで作製したMPO遺伝子欠損マウスを用いて解析した。その結果、MPOがMPO-ANCA産生の主要な抗原の一つになり、MPO-ANCAが血管炎進行に関与することを明らかにした。また、好中球異常を示す転写因子IRF-8/ICSBPの欠損マウスにおいて、好中球異常がMPO-ANCA産生に関与することを認め、MPO-ANCAは加齢とともに上昇することが判明した。一方、血管炎、腎炎の発症への好中球の関与を明らかにする目的で、半月体形成性糸球体腎炎を高率かつ急速に自然発症するSCG/Kjマウスを用い解析し、腎炎の発症、進行の初期段階(尿たんぱく300mg/dl以下)の末梢好中球が無刺激でMPOの易放出性を示し、活性化状態になっていることを示した。一方、LECT2は、肝細胞で高発現し肝炎などの肝細胞障害によりそのmRNA発現が低下した。現在、LECT2の機能解析が進行しており、LECT2は、血管炎にともなう多臓器不全の機構解析に有用な因子となると推定される。
