In vivo 脳虚血動物モデルにおける虚血抵抗性と脆弱性の法医学的研究

Research Abstract	平成13年度において、虚血負荷動物モデルを作成し、脳内微小透析膜法を応用した虚血再灌流後における側坐核及び視床下部の神経活動の抵抗性を比較検討した。微小透析膜プローブをガイドカニューレに沿って側坐核または視床下部へ挿入し2-3時間人工脊髄液灌流後のモノアミン放出の安定を確認した。その後、20分間40mMKCl含有灌流液に交換し、再び対照灌流液に戻した。1時間後再び同様の灌流液交換操作を行い、無麻酔下20分間隔で採取した側坐核及び視床下部灌流液中のドパミン(DA)、セロトニン(5-HT)をHPLC-ECDにより同時測定した。側坐核において、虚血処置群のDA放出は、Sham処置群と比較して、2回のK刺激により有意に増加した。側坐核5-HT放出には、虚血群及びSham処置群ともにK刺激により増加したが2群間の差は認められなかった。視床下部におけるK刺激DA及び5-HT放出は、増加を示したが虚血群、Sham群に差は認められなかった。視床下部は虚血再灌流障害に対して側坐核に比較して神経活動の抵抗性を示した。
