| Research Abstract |
生体内で最も強力な抗原提示細胞である樹状細胞(DC)がウイルス性肝炎や自己免疫性肝疾患、悪性腫瘍で機能異常があることを報告してきた。また、マウスから実際の病変の現場である肝内のDCの単離を行い、未成熟な樹状細胞であること、抗原を取り込んだ後に抗原特異的な免疫応答が誘導できることを明らかにした。
本年は、マウス肝炎モデルの作成とその際のDCの機能を解析した。肝炎マウスモデルは、C57BL/6マウスにConcancavalin A(Con A)を投与することにより作成した。このモデルでは、血清学的および組織学的に肝炎が起こっていることを確認した。コントロールとしてはPBSのみを投与した。肝炎発症早期の投与後8時間に肝樹状細胞を単離して、上記の性状と機能の解析を行い、コントロール群と比較検討した。肝炎群の肝樹状細胞の細胞表面マーカーの解析では、MHC-class II, CD86がコントロール群に比し、有意に増強していた。また、異型同種T細胞に対する増殖能も増強していた。一方、肝炎群では肝樹状細胞の抗原取り込み能はコントロール群に比し、低下していた。このことから、肝炎群では炎症によって非特異的な成熟がみられていることが示唆された。一方、抗原特異的な免疫応答を解析すると、肝炎群の肝樹状細胞はHBsAgとKLH特異的感作リンパ球に対する幼若化補助能が誘導できなかった。また、IL-2,IFN-yの産生誘導も有意に低下していた。すなわち、肝炎発症時には肝樹状細胞が免疫応答を誘導する能力が失われていることが明らかになった。
今回の検討から、肝炎ウイルスが肝内に侵入し肝炎が起こっている状態では、肝樹状細胞の抗原提示能の低下があるためにウイルスを排除する強い免疫応答が誘導できず、ウイルスキャリア状態になることが示唆された。
