Research Abstract |
TO INVESTIGATE THE BIOCHEMICAL MARKERS OF FETAL DISTRESS, WE EXAMINED NEUROTROPHIC FACRTORS IN AMNIOTIC FLUID FROM RATS RECEIVED TRANSIENT UTERIN ISCHEMIA. WE USED PREGNANCY 18 WISTER RATS, WHICH RIGHT UTERINE ARTERIS CRAMPED 30 MINUTES. AMNIOTIC FLUID OBTAINED BY AMNIOCENTESIS AFTER 24 HRS FROM CRAMPED. FOR EACH EXPERIMENT THE AMNIOTIC FLUID IN THE RIGHT HORN SERVED AS A ISCHEMIA GROUP AND THOSE IN THE LEFT HORN WERE USED AS A CONTROL GROUP. MEAHWHILE, WE PERFORMED IN VITRO EXPERIMENT FOR HYPOXIA. WE USED HUMAN AMNIOTC EPITHELAL CELLS DERIVED FROM PLACENTA BY CAESAREAN SECTION. CONTROL GROUP CULTURED IN 5% C02 INCUBATOR AT 37^<0>C FOR 48 HRS, ISCHEMIA GROUP INCUBATED IN 2% O_<2>, 5% CO_<2> INCUBATOR 37^<0> : FOR 48 HRS. STATISTICAL ANAYSIS BY WILCOXON SIGNED-RANK TEST DEMONSTRATED A SIGNIFICANT INCREASE IN NT-3 CONCENTRATIONS IN AMNIOTIC FLUID FROM THE ISHCEMIC GROUP IN CONTRAST NGF AND BDNF CONCENTRATIONS WHICH ALSO INCREASED BUT NOT SIGNIFICANT. IN VITRO EXPERIMNT SHOWED THAT NT-3 IN CULTURED MEDIUM OF HYPOXIC CELLS INCREASED SIGNIFICATLY. THESE RESULTS INDICATE THAT NT-3 IN AMNIOTIC FLUID MAY BE USED AS ONE OF BIOCHEMICAL MARKERS FOR FETAL DISTERSS.
