レプチン過剰発現トランスジェニックマウスを用いたレプチン抵抗性の分子機構の解明

Research Abstract	現在、アメリカ合衆国において肥満者に対するリコンビナント・レプチンの臨床トライアルが進行中であるが、大部分の肥満者が高レプチン血症を伴う「レプチン抵抗性」状態にあると考えられるため、末梢からのレプチン投与が充分に奏功しない事例が少なからず報告されている。本研究はこのような実情を踏まえて、申請者らが独自に開発したレプチンのトランスジェニックマウスがレプチン高感受性マウスであることに着目し、そのマウスを用いて「レプチン抵抗性」発症の分子機構を明らかにすることを目的に検討を行っている。昨年、レプチントランスジェニックマウスに高カロリー食を与えることにより、このマウスに肥満を誘導できることを新たに見出し、また、食餌中の脂質の含有量によって、レプチン抵抗性の発生機序が異なる可能性を考慮していた。今回、ob/obマウス、コントロールマウスそしてレプチントランスジェニックマウスをそれぞれ掛け合せて、レプチン血中濃度の異なる状況を作成し、高脂肪食負荷による肥満の誘導の程度を観察した。その結果、血中レプチン濃度の高低にかかわらず、高脂肪食で飼育した場合には同様に肥満が進行していくことが観察された。従って、現在の所、高脂肪食は、レプチン抵抗性とは無関係に肥満を誘導する可能性を考えている。しかしながら、高脂肪食による肥満誘導と同時にもたらされるインスリン抵抗性に対して、レプチンが生理的意義を発揮している可能性が残されており、今後インスリン抵抗性の程度など検討していく予定である。
