Research Abstract

1) We observed the effect of growth hormone and thyroid hormone using rat femur organ culture. Our studies indicate that the effect of growth hormone and thyroid hormone as local factors might have the reaction through some growth factors other than direct effect.
2) Callus formation is important on fracture healing. To investigate the effect of fixation instability on the hard and soft callus, the morphological changes in fracture callus were studied using a rat femur fracture model. A stainless steal pin or a nylon filament was used for internal fixation, and compared this to a group without fixation. Proliferating cells and TGF-β1 expression in fracture callus were examined by immunohistochemistry. Our studies suggest that greater instability causes more connective tissue generation associated with vigorous cell proliferation, resulting in enlarged cartilage tissue. However, excessive inter fragmentary movement at the fracture site suppress chondrocyte differentiation, and this is associated with predominant TGF-β1 expression.
3) To investigate the effect of growth factors on osteophytes in osteoarthritis (OA), we observed the expression of growth factors by RT-PCR and immunohistochemistry using osteophytes on femoral head during operation for total hip arthroplasty. We also used the femoral head cancellous bone with osteoarthritis or aseptic necrosis (AN) as the sample. TGF-β1 mRNA expression was seen in the surface of osteophytes and connective tissues intracellularly. B-FGF mRNA expression was seen in the deeper layer of osteophytes. On the other hand, expression was not seen in cancellous bone of OA and AN. B-FGF expression was more detected than TGF-β1 in the cartilage with degeneration in OA. These results suggest that TGF-β1 and b-FGF are involved in the formation of osteophytes. Different localization indicates that TGF-β1 seem to have stronger influence.
