Research Abstract

At first. the bioresorbable cement discs (5mm in diameter, 3mm thickness, 2.3 mm inside diameter) were soaked with 5μg of rhBMP-2 and dripped bone mallow blood (BMB) into the inner space. four kinds of discs.
1) added with rhBMP-2 and BMB. 2) added with rhBMP-2, 3) added with BMB, 4) only cement, were implanted into the subcutaneous tissue of SD rats After 3 weeks. the group 1) induced 3.5 fold of new bone than that of the group even at the center of the inner cavity.
Secondly, the cement cylinders loaded with 1.26 μg of rhBMP-2 were implanted into 5 mm bone defects of SD rat femurs. Using 18G needle, the bone mallow (BM) in contact with the cylinder was injected into the cylinder inner cavity. Three groups. 1) rhBMP-2+BM. 2) rhBMP-2, 3) BM, were compared. The group 1) united 60% and showed 70% of strength of the intact bone at 9 weeks. However, the group 2) united only 40% and the group 3) did not unite at 9 weeks.
At last, 5mm cement discs soaked with 5 μg of rhBMP-2 and loaded with 10^4 cultured bone mallow cells (BMC) were implanted into dorsal subcutaneous tissue of Fisher rats. Six groups. 1) rhBMP-2+BMC, 2) rhBMP-2, 3) BMC. 4) collagen disc+rhBMP-2, 5) collagen disc. 6) cement alone, were compared. At. 6 weeks, the group 1) induced more matured bone and two fold quantity of new bone than that of the group 2) However. other groups did notinduce new bone.
