卵巣癌細胞の増殖能及び浸潤能に関与するプロテアーゼ遺伝子の同定と治療有用性の検討

Research Abstract

本研究は卵巣癌細胞で過剰な発現を呈する既知および未知のプロテアーゼ遺伝子を同定し、それらが卵巣癌の治療に応用され得る可能性についても検討することを目的とする。
平成13年度はDifferential Displayとそれに続くサブクローニングにより卵巣癌細胞において過剰発現を呈するセリンプロテアーゼ遺伝子として同定されたStratum Corneum Chymotryptic Enzyme (SCCE)及びその特異的インヒビターであるAntileukoprotease (ALP)の発現について検討し、SCCEは卵巣癌の進展に関与するプロテアーゼの一つであり、その機能調節にはALPが関与している可能性があることを報告した(Int. J Gynecol. Cancer 11:454-461,2001)。さらにメタロプロテアーゼ発現異常に関する研究成果をMatrix Metalloproteinases and Ovarian Cancer (Recent Research Development in Cancer, Transworld Research Network, Trivandrum, pp283-293,2000)として出版した。さらに、それらのプロテアーゼ遺伝子異常と、細胞周期調節因子異常の関連を検索する目的で、卵巣腫瘍におけるp53、p27、cyclin Eの異常について検討し、それらの成績を報告した(Int. J. Oncol. 18:953-958,2001、J. Soc. Gynecol. Invest. 8:179-185,2001)。
現在、我々はいくつか新規プロテアーゼ遺伝子のクローニングも行っており、卵巣癌の増殖、浸潤、転移における一連の複雑な過程におけるプロテアーゼの役割の解明を進めている。
