| Research Abstract |
1)炎症・発痛モデル動物(ニューロパチックペインモデル)の作成
SD雄性ラットを用いCCI(Chronic Constriction Injury)モデルを作成した。その後、疼痛関連行動にて動物行動学的に評価・解析し、ニューロパチックペインモデル動物を作成した。
2)ニューロパチックペイン発症時の中枢での興奮性アミノ酸関与の解析
脊髄後角・後索核での神経情報伝達に興奮性アミノ酸であるグルタミン酸がどのように関与しているか各種グルタミン酸受容体拮抗薬を用いて薬理学的、電気生理学的に検索した。またグルタミン酸受容体の遺伝子発現を形態学的に解析した。
a)薬理学的検討
AP-5,MK-801,CNQXなどの各種イオンチャンネル型グルタミン酸受容体拮抗薬をBath Application、Intra Venousなどの方法で投与し特にevoked response等の反応にを検索した。
b)電気生理学的検討
上記薬物投与に対する影響を特にWide Dynamic Range (WDR) neuronに関する細胞外単一神経活動電位の記録によって解析した。
c)形態学的検討
モデル動物より新鮮凍結切片を作製し、RI標識したoligonucleotide probeにて組織上のグルタミン酸受容体の遺伝子発現(mRNA)を解析した。
3)データ構築とまとめ
上記のデータをまとめ論文作成、学会発表をおこなった。
