Research Abstract |
The elimination mechanism of ghost tangle was observed immunohistochemically and immunoelectron microscopically in the CA1 of the parkinsonism-dementia complex of Guam. Neurofibrillary tangles in the neurons were mainly composed of pared helical filaments (PHFs), but the ghost tangles were of straight tubules (STs). Astrocytic processes were tightly intertwined with ghost tangles in immunohistochemistry. Immunoelectronmicroscopically, astrocytic glial filaments were admixed with the ghost tangle STs. However, microglial participation was not remarkable. Neurofibrillary tangles, including PHFs and STs, were never observed in the lysosomes. The findings indicate that the ghost tangles were eliminated by the astrocytes. However, the plasma membrane of the astrocytes were not clear in these observations due to postmortem autolytic alteration. Thus the practical sites, that is, inside of astrocytes or in the extracellular space, were not clear in ghost tangle elimination. To overcome the problem, experimental study is inevitable using animals and NFT inoculation to the brains.
