| Research Abstract |
昨年度構築したインスレーター活性を検出するためのレポータープラスミドpTIA(tester of insulator activity)を用いて、マウスゲノム断片からインスレーター活性を含むものを単離することを試みた。しかしながら、ゲノム断片挿入によるスペーサー効果と明瞭に区別しうるインスレーター活性の検出には至っていない。この過程で、他施設からの報告により、インスレーター配列のエンハンサー活性遮断効果は、当該エンハンサーの両側にインスレーターがタンデムリピートとして配置されることにより増強することが明らかになったので、現在これを踏まえたベクターデザインの改良を検討している。
一方、昨年度の検討でES細胞においてインスレーター活性が検出できたニワトリβ-globin LCR(locus control region)由来CTCF結合配列を用いたインスレーターカセットに、比較的強力な活性を示すhuman β-actin promoterないしは極めて弱い活性しか示さないhCMV^*-1 promotorの制御下にβ-geo(β-galactosidase+neomycin耐性遺伝子の融合蛋白をコードする)を接続したレポーターカセットを組み込んで、これらをES細胞に導入した。この結果、インスレーターは弱いプロモーターがゲノム上の挿入部位近傍のエンハンサーから受ける活性化は遮断できるが、クロマチン構造に起因すると考えられるプロモーター活性への抑制効果は遮断できないと考えられた。今後、これらの結果をさらに種々の異なる方法で検討するとともに、より有用な外来遺伝子発現のためのカセットの構築を進めていきたい。
