Research Abstract	これまでに行われてきたDNA分子の質量分析では、測定における種々のパラメータのうち、どのパラメータがどのように測定値に影響するのかという点に関して十分な検証がなされていなかったため、再現性の高い結果を得ることは困難であった。そこで我々は、まずはじめに(1)DNA鎖長、(2)DNA濃度、(3)DNA溶液の純度、という3つのパラメータに着目し、詳細な条件検討を行った。検討の結果、最適な条件を見出すことに成功し、当初20〜50bpまでのDNA分子の検出が限界であったのに対し、現在では200bp程度まで極めて再現性良く測定することが可能となった。それと同時に、飛躍的な分解能の向上を実現した。一方、ハード面では、次世代のMALDI-TOF質量分析計であるAXIMA-CFRを新たに開発した。本装置は、デフォルトの設定では384検体の質量分析を一回の測定で行うことが可能であり、フリーフォーマットでのサンプルウェル指定を行えば数千検体の処理を一回で行うことが可能である。このハイスループット性は、今後本研究を実用化する際の重要な要素として位置付けられる。現在は、マトリックスの検討も含め更に詳細に種々の条件検討を行っている。それに加え、質量分析計の更なる性能の向上に取り組んでいる。これらのソフト面、ハード面の両方向からのアプローチにより、500bp程度のDNA分子についての安定した測定系を確立し、最終的には研究面・臨床面での実用性・応用性に十分に耐え得るDNAチップを作製したいと考えている。
