| Research Abstract |
1 高頻度で長い距離の異種間形質転換を生じさせる突然変異体分離のための菌株を育種した。
Bacillus subtilis AWG2(cysA metC hisH leuB arg-1 aroG ist1)株を育種した。この菌株は異種間で顕著に組換えを起こすマーカーを多重に持つ。
2 突然変異体を分離した。
B. subtilis AWG2(cysA metC hisH leuB arg-1 aroG ist1)株をN-メチル-N'-ニトロ-N-ニトロ-N-ニトロソグアニジンで変異処理した。処理細胞からコンピテント細胞を調製し、B. amyloliquefaciens FWSの染色体DNAによるArg^+Aro^+同時形質転換体を分離した。その中から、高頻度にMet^+異種間形質転換を行う変異体を分離した。その変異をist2変異と名付けた。
3 突然変異体の特徴づけを行った。
ist1 ist2変異体は,異種の壁を大いに乗り越えられる際だった特徴を示した。
(1)異種菌(B. amyloliquefaciens FWS)の34kb領域を一気に組み換えた。長い距離の異種間組み換え。
(2)B. megateriumやB. haloduransのDNAのcysEからrpsL領域の17kbをも組み換えた。特にB. haloduransの場合DNAの相同性は63%しかなく、異種の壁を大いに乗り越えらる組み換え。
4 遺伝子マッピングを行う。
細胞融合法、PBS1ファージ形質導入法で遺伝子領域を限定した。ist1はyfhDとmetCの間、ist2はpyrDの近傍。PCR法などを用いて遺伝子特定を行っている。
5 平成13年度総括
平成13年度の当初の目的はほぼ達成できた。また、高頻度形質転換法の特徴を公表した。種の壁を大いに乗り越えられる方法を認めた。
