Research Abstract	1)グリア細胞に存在する水チャネルアクアポリン-4の構造を解析するために、昆虫細胞Sf9を用いて大量発現し、精製した。Sf9細胞の6リッターカルチャーから10mgのアクアポリン-4が精製できるようになったので、これを用いて2次元結晶を作製する条件を検討できるようになった。その結果、3Åを超える分解能の電子線解説図形が撮影できるような2次元結晶が作製できた。 2)電気エイの発電器官から作製されたニコチン性アセチルコリン受容体(nAChR)のチューブ状結晶を極低温電子顕微鏡で撮影して、立体構造解析を行って、nAChRの4.5Å分解能の解析とX線結晶学により解析されたアセチルコリン結合蛋白質の分子構造の比較から、リガンド結合に伴い、この受容体の2つのαサブユニットの内側のβバレルが15度程度回転するという構造変化の機構を解明したが、引き続き分解能を向上させて、4Å分解能での解析から、原子モデルを作製した。この結果はNatureに(Articleとして)投稿した。 3)IP_3受容体の立体構造を、極低温電子顕微鏡と単粒子解析法を用いて解析した。 4)ギャップジャンクションにおいて、聴覚障害の原因として知られるコネキシン26の変異体の解析を行い、J.Biol.Chem.に発表した。 5)ヒトエンドセリン受容体B型とカベオリン1との相互作用を研究して、ヒトエンドセリン受容体B型におけるシグナル伝達の制御機構を解明して、EJBに発表した。
