| Research Abstract |
山口はショウジョウバエDNA複製関連遺伝子の転写制御エレメント、URE配列とDRE配列の間に存在する新規の転写調節エレメント(PRE配列)を見出し、それを標的とした酵母one-hybridスクリーニングを行い、新規の転写因子のcDNAをクローン化した。この因子はRFXドメインを持つことからdRFX2と名付けた。またこの因子のDNA結合ドメインをショウジョウバエ複眼原基特異的に発現させると複眼形態異常をもたらすことがわかった。また、ショウジョウバエDNA複製関連遺伝子を共通に制御する転写因子DREFと相互作用する因子としてホメオドメインタンパクDistal-lessとクロマチンリモデリング因子Mi-2が新たに同定された。DREFをショウジョウバエの複眼原基で過剰発現させると、成虫の複眼が形態異常を示すが、この遺伝子導入ハエ系統とDistal-less突然変異系統あるいはMi-2突然変異系統とを交配すると、この形態異常を増強することから、Distal-lessとMi-2はDREF機能に抑制的に働いていることが示唆された。
松崎はMirandaの局在を指標に、ショウジョウバエ神経幹細胞の非対称分裂に異常をきたす致死突然変異を系統的にスクリーニングし、数種類の責任遺伝子を同定した。その中には、がん抑制遺伝子や、細胞分裂期の収縮環形成に関与する遺伝子等が含まれている。
