Research Abstract	本研究班は平成13年度に発足し、平成17年度までに、我々の体を構成する部品である器官(臓器)の形成に関わる遺伝子プログラムと、器官を構成する組織間の相互作用の実体を明らかにすることを目的としている。この研究は、人工臓器や器官再生といった社会的要請にも基盤的知見を提供するものである。平成15年度に新たに得られた主要な成果は以下の通りである。(1)ニワトリ胚消化管の発生においては、繊維芽細胞成長因子(FGF)が重要な役割を担っていることが明らかになった。FGFは胃の上皮細胞の分化形質に影響を与え、その過剰発現は胃腺上皮の縮小と内腔上皮の拡大をもたらす。また、胃腺上皮細胞におけるペプシノゲン遺伝子の特異的発現について、その調節領域と関係する転写因子が明瞭に同定された。(八杉)(2)プラナリアの再生に際して、成長因子Wntとその関連遺伝子が重要であることが示された。またNotchシグナル関連遺伝子についても、Notch,Delta,sno,Su(H)遺伝子などが次々とクローニングされ、RNAiなどの先端的方法を用いてこれら遺伝子の機能が解析された。(阿形)(3)アフリカツメガエルのアニマルキャップを用いる系を駆使して、膵臓、肝臓などの機能的な臓器を創出し、そこで作用する遺伝子の時間的、空間的発現が詳細に調べられた。これらの器官形成においては、遺伝子が正常発生と同様のパターンで発現することから、この系は正常発生の研究の優れたモデルになるものである。(浅島)(4)ニワトリとヒトのiroquois遺伝子の、脳の分化における機能が解析され、FGFシグナルとの関係が明らかにされた。また、これまでにも追求されてきたTbx遺伝子の心臓発生における役割が一層詳細に解析され、心室や心室中隔の形成における重要性が示された。(小椋)(5)ニワトリの肺芽発生にFGFが関与していることが実験発生学および分子生物学的に示された。またFGFの発現はTbxによって正または負に制御されること、肺芽の出現位置の決定にはHoxb-6が関与することが示唆された。(黒岩)(6)ショウジョウバエは多くの知見を与えるモデル動物である。その翅と視神経の投射におけるシグナル伝達機構が解析された。ここで重要なのはdppやWntであり、これらの因子の機能亢進や機能阻害は種々の奇形、機能不全を引き超した。(多羽田)以上のように、平成15年度においては、本来の目標である器官形成におけるシグナル伝達機構のまさに中心に切り込み、かつ、種々のシステムで時としてよく類似したシグナル伝達系が内包されていることを明らかにした。
