| Research Abstract |
本研究では、(1)アジアの中の日本という立場からアジアのヒトゲノム多様性研究、(2)ヒトに焦点を当てた進化研究のための霊長類ゲノム研究、に資することを目的とし、「ヒト及び霊長類の細胞・遺伝子レポジトリー」の確立を目指す。
遺伝子試料の収集と細胞株の樹立については、本年度は倫理指針の関係からヒトに関しては新たな試料収集はおこなわず、現在1900株ある樹立細胞の整備をおこなった。霊長類に関しては、樹立細胞株は、類人猿から原猿に至る29種、174株になった。
汎用株化法の確立については、EBウイルス法のマニュアルを作成し、使い勝手のモニタリングと、汎用細胞株化法の研修をおこなっている。現在、ウェブサイトで3カ国語(和・英・華)で公開すべく準備を進めている。
低コスト化の視点から細胞株化技術の改良については、1検体あたり200ドルのところを75ドル近くまでコストダウンできた。
使用可能となった試料を用いた研究で、SDF1-3'Aのアジア・メラネシアにおける分布の人類学的背景,AIM遺伝子多型と3大人種の関係,mclrの多型分布と皮膚色・淘汰説の関係が明らかになった。また、霊長類の試料を用い、いくつかのヒトの多型の起源を明らかにした。現在5研究機関に試料を提供し共同研究が進められている。
