転写統合装置複合体機能調節による乳癌発症機構の解析

Research Abstract

これまで、申請者は本研究テーマに関して以下の成果を得た。
(1)RNAヘリケースA(RHA)/CREB結合タンパク質(CBP)複合体の発見(Cell 1997,G＆D 1997)
(2)BRCA1との関連(Nature Genetics 1998)
(3)同複合体の神異変性症(細胞死)への関与(Nature Genetics 2000)
(4)同複合体のβ-cateninによるアポトーシス回避機構(JBC 2000)
(5)RHAによる2種類の転写活性化機構の証明(MCB 2001)を証明した。
(5)に関しては、CREBの転写活性化にはATPase/ヘリケース活性、およびRNA polymerase II複合体のリクルートメント活性の両者が必要であるが、BRCA1の転写活性化には、ATPase/ヘリケース活性よりもpol II複合体のリクルートメント活性の方が、より重要であることが明らかとなった(ArataniらMCB 2001および投稿準備中)。この点では、BRCTとよばれるBRCA1のC端に存在する転写活性化領域とRHAが結合し、その活性化に必要であるとの以前のわたしたちの報告(Nature Genetics 1998)と合致するものである。
また、現在、(6)細胞周期関連因子、ガン抑制遺伝子産物がRHA複合体に含まれ、かつ細胞周期の調節に関与していることが明らかとなっている。
