| Research Abstract |
私どもは肝幹細胞を移植することにより、重症肝不全に対する抜本的治療法の開発を目指している。具体的にはMusashi遺伝子(Msi-1)をマーカーとして文字通りの肝幹細胞の単離を実現させ、病態に応じた戦略的治療法を開発レようとレている。
平成13年度において、ラット肝幹細胞であるoval cell lineを樹立し、この細胞が肝細胞と胆管上皮細胞の両方の形質を併せ持つことを確認した。さらに、この幹細胞がMsi-1遺伝子を発現しており、スフェロイド化により肝細胞に向けて分化させると、Msi-1遺伝子の発現が抑制されることを見出した。さらにヒト肝がん由来細胞株7株のうち、3株にMsi-1遺伝子が発現していることを見出した。このことはMsi-1が肝幹細胞の分化や肝発がんに係わっていることを示唆している(Biochem Biophys Res Commun 293:150-154,2002)。
平成14年度において、oval cellsにおけるMsi-1遺伝子発現はNmb遺伝子発現抑制を介してノッチシグナルを制御していることを明らかにした。また、Msi-1遺伝子ノックアウトマウスを用いて、肝発生に関するMsi-1の役割について検索中である。
