| Research Abstract |
これまでの研究から第14番染色体での癌抑制遺伝子としての候補遺伝子を詳細に解析してきたが現在のところ急速進行性腎細胞癌において特異的に発現の低下を認める遺伝子座は同定できていない。
しかしこれらの遺伝子座でのメチル化による修飾を検討する中で腎細胞癌では複数の癌抑制遺伝子が不活化していることが判明した(Kawakami et al.,Urology 2003)。
特にP16,p15といった第9番染色体上の遺伝子群と第16番染色体上のE-cadherin遺伝子が同時に不活化している例が高頻度に存在し、しかもこの不活化のプロセスとしてはp16,p15といった第9番染色体遺伝子ではhomozygous deletion、E-cadherin遺伝子については遺伝子欠失を伴わない両側対立遺伝子のメチル化というようにそれぞれの遺伝子が特異的な不活化機構を有することがわかってきた(Oncogene 投稿中)。
現在第14番染色体長腕上に存在する急速進行性腎細胞癌の責任遺伝子同定には至っていないが初年度に見出した候補遺伝子IkappaBのExon4,5,6の多型とparaneoplastic syndromeの発症との関係をSNPとしての病態感受性とのかかわりについても検討していきたいと考えている。
