| Research Abstract |
(実績1),タコ皮膚から光受容体の単離精製
タコの皮膚の色覚色素を含む膜断片をタコ皮膚から蔗糖密度勾配法により精製後界面活性剤で可溶化し、その抽出液の分光学的性質を調べた。照射波長により吸収スペクトルの可逆的変化が見られる光受容体が存在した。この抽出液を分子フルイで分画したところ、分子量1万以下の溶液に大部分の光受容体が存在することを確認した。分子量からオモクロームを持った色素タンパク質であると考えられる。レチナールタンパク質は分子量4万以上と考えられ、分子フルイで分子量4万以上の分画に注目し、イオン交換カラム等で精製をしている。
(実績2)タコ皮膚の色覚色素の遺伝子クローニング
ノザーンブロッティング解析においてタコロドプシン遺伝子と交差するバンドを得ている。すでに得たタコ皮膚のcDNAライブラリーではポジティブクローンを得られなかったので、現在新たにタコ皮膚のcDNAライブラリーを作成している。
