Research Abstract	インターフェロン・タウ(INFτ)遺伝子の低濃度・持続性発現のために、INFτ遺伝子の上流域の解析を引き続き行った。当初、この解析にヒト絨毛性ガン細胞培養系を用いながら行った結果、プロモーター領域よりもエンハンサー領域に結合する核タンパクAP-1が、INFt遺伝子の発現を制御していることが分かった。しかし、シバヤギのトロホブラスト細胞株で解析すると、プロモーター領域のEts-2がより重要なことが判明した。さらに、コアクチベーターCBPの関与も証明したが(Xu et al. Mol. Reprod. Dev.2003,In Press)、低濃度・持続的発現を実現するまでには至らなかった。そこで、フォーミーウイルス(FV)遺伝子の発現・導入ベクターを開発し、INFτ遺伝子を導入した、いわゆるLive Vectorの開発とその評価系を確立した。すなわち、INFτ-ヒトFV(Live Vector)をマクロファージ細胞にトランジェント、スティブル・トランスフェクション法にて導入し、INFτの発現を確認した。その後、nef領域にGFPを導入したHIVプラスミドを構築し、それをHeLa細胞に感染させることによってHIVのウイルス粒子を産生した。そのウイルス粒子を先のINFτ発現細胞に感染させたところ、HIVの感染を効率的に減少させることができた(JVMSに投稿、現在改訂中)。そのため、INFτ-FVはLive Vectorとして有用性が高いことが判明した。現在、INFτの発現強度とHIVの感染防御について検討中である。
