We performed a case-control study to clarify the relationship between the formation of Helicobacter pylori (H. pylori) infection and polymorphisms of immune-related gene from April 2001 to March 2004. Study participants were healthy Japanese, who attended for annual medical check-up. written informed consents were obtained by all subjects for this study, which was approved by ethical committee of Shimane University. we selected 96 subjects with seropositive of H. pylori and age-and sex-matched 96 subjects with seronegative of H. pylori. Genomic DNA was extracted from white blood cell and analyzed the genomic polymorphisms in promotor region (-308) of tumor necrosis factor (TNF)-α, IL-10, CD 14 and two sites of Mannose-binding lectin (MBL). As a result, meaningful differences in polymorphism of five immune-related genes, we investigated in this study, were not observed between H pylori-seropositive and seronegative subjects. Therefore, we concluded that the difference in hereditary immune mechanism may not influence the formation of H. pylori infect ion.
During we performed this study, we clarified chat long-term H. pylori infect ion reduces serum level of high-density lipoprotein cholesterol. However, the degree of arteriosclerosis is influenced by aging and not by H. pylori infect ion. In addition, serum homocysteine level is not different between H. pylori-seropositive and negative individuals. We reported these findings in English medical journal.
