Research Abstract	■マウス結核感染症に対するDCを用いたvaccineの有効性を検討するために,以下の抗結核vaccineを作製した. 1)結核菌由来の抗原ペプチド(Ag85A,Ag85B)をcodeしたDNA vaccine 2)結核菌由来の抗原ペプチド(Ag85A,Ag85B)を遺伝子導入したDC Ag85A,Bを発現させたplasmidを用いたDNA vaccineを作製し,また,これらのAg85A,BのcDNAを挿入したpMX vectorをretrovirus-producer cell(Phoenix cell)にtransfectionして培養液からretrovirusを回収し,マウス骨髄由来の培養DC(BALB/c,C57BL)に感染させて遺伝子導入を行った. 各種vaccineによるマウスの免疫は,DNA vaccineは金粒子に付着させて,遺伝子銃で皮内に接種,遺伝子導入したDCは静注,DC exosomeは皮内に接種して行った.
