Research Abstract |
In cord blood transplantation, graft rejection is one of the major concerns, especially in non-malignancy. There are several cell types that are closely related to graft rejection. Dendritic cells (DC) present allogeneic peptide to T cells that attack host. DC are subclassified into two types, type 1 and type 2. In this project, we examined whether DC may enhance allo-reaction. Since our preliminary data showed that DC could be induced from cord blood monocytes, we examined the characteristics of cord blood monocytes. Cord blood monocytes showed lower antigen-presenting capacity than adult blood, and functionally immature (Pediatr Int, 2001 and Clin & Exp Immunol 2001). Next, we have analyzed what kind of molecules are associated with DC function. We found that Rho GTPase plays a key role in the regulation of morphology and function (J Immunol, 2001). Then, we have investigated the G-protein-coupled receptors (GPCRs) in monocytes and have found that LSSIG is induced by the activation of STAT3 (Blood, 2003). Taken together, these results suggest that DC and its precursor (monocytes) can be used to regulate graft rejection.
