Research Abstract

(1)BALB/C mouse(Th 2 type)とB/6 mouse(Th 1 type)を抗原とStaphylococcus aureusの膜成分をFreund incomplete adjuvantと共に2週毎に皮下に免疫した.免疫後4、8、12、16,24週目に屠殺した.
(a)BALB/C mouseでは周齢が高くなるにつれ、糸球体メサンギウムにIgG,IgA,C3抗体、ならびにStaphylococcus aureus抗原の沈着が増加した.(b)産生された抗体をヒトHGGでprotein Aで吸収した膜抗原をcoateしたELISA法で測定したところ、対照群に比して有意に抗Staphylococcus aureus抗体(IgGおよびIgA)の上昇をみた.c光顕では軽度〜中等度のメサンギウム増殖性腎炎が発症した.(d)一方対照群のB/6群では軽度の免疫グロブリンの沈着を認めるも、Staphylococcus aureus抗原の局在は認めなかった.(e)BALB/c mouse群では対照群に比して、有意に多量の蛋白尿を認めた.(f)BALB/mouse免疫後の脾細胞にStaphylococcus aureus抗原を加え、培養したところ、有意に抗Staphylococcus aureus IgA抗体の上昇を認めた.
(2)抗Staphylococcus aureusに対するmouse monoclonal antibodyの作成:PG法によりBALB/Cの脾細胞とNS1とhybridomaを作成し、モノクローナル抗体を得た.この抗体を用いて、ヒトIgA腎症患者の組織中のStaphylococcus aureus抗原を確認すると同時に、Staphylococcus aureus菌体成分に対するwestern blotを行い、35kDのバンドと反応することを確認した.
(3)抗Staphylococcus aureus抗体を用いて、免疫沈降法で得られた抗原のアミノ酸配列を検討し、Staphylococcus aureus膜成分の一部であることを確認した.
以上の結果から、Staphylococcus aureus膜抗原を用いたmouse IgA腎症類似病変の惹起には、Th 2 typeの免疫遺伝学的背景を有することが重要である.今後IgAへのクラススイッチ機序等を検討する予定である.
