血管平滑筋細胞におけるエリスロポエチンの情報伝達に関する検討

| Research Abstract |
平成13年度は,Epoの血管平滑筋細胞における作用を明らかにするために,EpoのMMPs産生及び活性化に対する影響について検討する予定であったが,結果はnegativeだった。従って,Epoの血管平滑筋細胞の増殖刺激作用に対するcAMP-protein kinase A系の影響を,Epoの情報伝達の各stepにおいて検討した。
結果はrHuEpo添加によりDNA合成は促進した。cAMP刺激薬のForskolinの前処置により,DNA合成は抑制されたが,cAMPのantagonistであるRp-cAMPによりその抑制は相殺された。
rHuEpo添加によりElk-1のリン酸化の亢進が時間依存性に認められ,そのpeakは5minであった。FKの前処置によりElk-1のリン酸化は抑制され,Rp-cAMPにより抑制は解除された。MEK, MAPKそれぞれのリン酸化もrHuEpoにより促進されるが,FKによるリン酸化の抑制を認めた。Protein kinase Cに依存するRaf-1-MEK-MAPKの活性化と,cAMPによる活性化抑制の関係を検討した。PKCのactivatorであるphorbol 12-myristate 13-acetate (PMA)添加によりMAPKのリン酸化の増強が認められた。FKの前処置によりPMAによるMAPKのリン酸化は抑制された。
結論としてVSMCにおいてcAMPの上昇は,主にPKCによるRaf-1の活性化を阻害することで,rHuEpo誘導性のRaf-MEK-MAPK系の活性化を阻害する可能性が示唆された。cAMPを上昇させる抗血小板薬の,Epo誘発性血圧上昇の抑制効果の一部は,この機序による可能性がある。
