ヒト下垂体腺腫におけるPACAP・PVRmRNA発現とアポトーシス調節機構

Research Abstract	申請者は下垂体腺腫においてPACAPおよびその受容体mRNA発現を検討し、腺腫型によりそれらの発現が異なり下垂体腺腫の機能上PACAPが選択的調節を行っていることを報告した(Am J Pathol 153:1787-1796,1998)。今回の申請では下垂体腺腫組織においてapoptosisの調節にcyclin,cyclin dependent kinase(CDK)およびそれらの調節因子であるp16,p21,p27,p53の発現を検討している。平成14年度では手術時に摘出されたヒト下垂体腺腫組織の凍結切片を用い、TUNEL法でapoptosis indexを算出し、同様の組織を使用しPACAPおよびその受容体遺伝子であるPVR-1,2,3mRNAを検出した。まず、TUNEL法によるapoptosis indexは24検体の下垂体腺腫(非機能性下垂体腺腫19例、成長ホルモン産生下垂体腺腫5例)では非機能性腺腫では2.5%、成長ホルモン産生腺腫では2.7%と腫瘍型による差は見られなかった。また、PVR mRNAsの発現はRT-PCR法により両腫瘍型でもPVR-1,PVR-2,PVR-3 mRNAの発現を認め、特にPVR-2 mRNAの発現が強いことが示唆された。平成15年度ではPACAPおよびPVR1〜3mRNAsの検出を通常のin situ hybridization等で試みたが発現量の問題から検出感度が低いことが想定され、現在最近当施設に入ったreal time PCRを用い検討している。その結果、PACAPは下垂体腺腫内にはほとんど存在せず、PVRは種々の頻度で下垂体腺腫内に認めた。p16,p21,27の発現は下垂体腺腫に認められるがp53の発現ではほとんど観察されなかった。
