Research Abstract |
1)Primary culture of human and rat meniscus-derived cells and mesenchymal stem cells Meniscus derived cells were harvested from excised meniscus and mesenchymal stem cells were also extracted from bone mallow of iliac bone and tibia. These cells were introduced to the primary culture and were analyzed for the production of extracellular matrices. The analysis of gene expression revealed that human meniscus cells and rat meniscus cells produced type I, II, and III collagen genes and cartilage proteoglycan gene. 2)Three-dimensional culture using collagen scaffold The primary culture of the human meniscus derived cells were seeded onto collagen scaffold and were maintained for four weeks. These cells attached to the collagen scaffold and produced the extracelluar matrix. 3) Biomechanical simulation during in vitro cell culture Biomechanical stimulation of cyclic axial loading to the three-dimensional culture of human mesenchymal stem cells effected cellular proliferation.
