Research Abstract

Our purpose is to investigate the effectiveness of radial artery. Human radial artery was obtained from patients undergoing coronary artery bypass grafting. The vessels were prepared and 3 mm rings suspended on wires in 20 ml organ chambers filled with buffered salt solution maintained at 37 ℃ and aerated with a mixture of 95% O2 : 5% CO2. One of the wires was anchored in the organ chamber and the her was connected to a force transducer. The preparations were repeatedly exposed to 60 mM of KCl to induce contraction at each level of stretching, until a maximal contractile response was obtained. We decided resting tension to 4g. The endothelial function was assessed by the rings to 10^<-5> M of acetylcholine. The endothelial function was not almost residural. Cumulative concentration-response curves for olprinone (10^<-9> up to 10^<-4>M) or nicorandil (10^<-9>up to 10^<-4>M) were obtained from tension measurements of rings contracted by norepinephrine after a pre incubation with propranolol (10 μ M). The vasodilator responses of olprinone or nicorandil were expressed as a percentage of the initial contraction induced by norepinephrine, and were dose dependent relaxant. Dose response curves to serotonin, U46619 or phenylephrine were obtained dose dependent relaxant by nicorandil, Relaxation dose response curve to nicorandil is examing in the presence of glibenclamide (10 μ M) or L-NAME (300 μ M) recently.
