Research Abstract

1)The establishment of diabetes mellitus model animal.
Of the medicine, Streptozotocin (STZ) which causes the diabetes mellitus to male SD strain rats of the 6〜8 week-old, 6mg/0.1ml/100g, it was injected into vein at the fraction of the body weight The blood glucose level of the animals rose 2 days after the, injection, and the albidus of the crystalline lens was recognized at the 2〜3 month, and the cataract was able to be induced. The macroscope findings were photographed by the newly purchased digital camera, and the results were preserved. The animals were fixed with 3% of paraformaldehyde by low pressure perfusion, and it was made to be post-experimental material that they were frozen or fixed in the alcohol, after retina and crystalline lens were extracted.
2)Experimental groups
Irsogladine malate (IM), 6.7μg/0.3ml/100g body weight of rat, was dissolved in water, and 1 time in every day was administered orally. Diet and water except for the medicine were taken ad libitum and prepared … More following.4 groups as experimental group.
(1)IM was administered every day for I week, then STZ was injected.
(2)STZ was once injected intravenously, then IM was administered every day for 1 week.
(3)STZ was once injected intravenously, then water was administered every day for 1 week.
(4)Physiological saline was once injected intravenously, then water was administered every day for 1 week.
3)Analysis and results
The ocular tissues such as crystalline lens or retina from the experimental or normal rats were made thin sections and they were stained with andi-connexin 32,43 or 50 antibodies. But it was difficult to make sections because of the mixed existance of soft and hard tissues. Now we are trying to make the sentions, therefor we did not get the clear results.
4)Other
The organs which was affected reconstitution of gap junction such as peripheral nerve, liver, kidney, lacrimal gland and so on, were examined. Apart of data was reported in J. Electron Microsc., Prog. Diabetes Research and Med. Electron Microsc. Less
