Research Abstract

To investigate the pathological mechanism of the development of odontogenic cyst and odontogenic tumor, the effects of carcinogens, Trp-P-2 and MeIQx, and growth factor TGF-α on the mouse tooth-germ in vitro and in vivo. In experiment I, effects of Trp-P-2 and TGF-α , alone and in combination, on the cell proliferation and differentiation of the organ-cultured molar tooth-germ of ddY embryonic mouse were studied histologically and immunohisochemically (PCNA). TGF-α increased the cell proliferation of the organ-cultured tooth-germ, but inhibited its differentiation. Trp-P-2 inhibited the cell proliferation of the organ-cultured tooth-germ. In experiment II, effects of MeIQx and TGF-α, alone and in combination, on the cell proliferation and differentiation of the mouse molar tooth-germs, which were transplanted into BALB/C nude mouse following organ-culture. Both of MeIQx and TGF-α increased the cell proliferation of the transplanted mouse tooth-germ. In experiment III, effects of Trp-P-2 and TGF-α, alone and in combination, on the cell proliferation and differentiation of the mouse molar tooth-germs, which were transplanted into BALB/C nude mouse following organ-culture. Trp-P-2 in combination with TGF-α increased the cell proliferation of the odontogenic epithelium of the transplanted tooth-germs, which showed keratinization and cyst formation. The results of the present study may suggest that some carcinogens and growth factor are involved in the development of odontogenic cyst and odontogenic tumor.
