Research Abstract

The DNA-lipid complexes with and without cytokines BMP and b-FGF were prepared synthetic lipids from the reaction of n-alkyl alcohol (C=4〜C=20) and amino acids (glycine, L-alanine and L-glutamic acids) and the DNA films were obtained by casting the DNA -lipid complexes from chloroform/ethanol solution. The tensile strengths and intercalation behavior of DNA films were investigated. Antibacterials characteristics of DNA-lipid complexes against bacteria (P. gingivalis, S. mutans, S. aureus and P. aeruginosa) were also investigated. Additionally, effect of the DNA -lipid complexes with cytokines BMP and b-FGF on bone formation was evaluated. The DNA-lipid films had tensile strengths of approximately 2MPa, and formed intercalation complexes with ethidium bromide. The formation of the intercalation complex of DNA-lipid film with ethidium bromide was influenced by the length of the alkyl chains of the synthesized lipids. The complexes prepared from DNA with glycine decyl ester or glycine dodecyl ester, and with L-alanine decyl ester or glycine dodecyl ester showed antibacterial activity. However, the DNA-lipid complexes with cytokines BMP and b-FGF did not elicit bone formation.
