Research Abstract |
This research has shown clearly that constitutively proteins are inducted at 37 ℃, 3 hours heat treatment, and that Hsp (heat shock proteins) of stress proteins (chaperon molecule) are inducted at 42℃, 3 hours in both a human gingival epithelium cell (HGK) and a human gingival fibroblast (HGF). After acting A.actinomycetemcomitans on HGK, expressions of stress proteins were analyzed by RT-PCR method and the western blotting method. Expression of Hsp27, Hsp60, and Hsp70 was recognized. These expressions were found in cell layer. As for Hsp90, expression was not accepted by heat treatment of 37℃ nor 42℃ at HGK. When Geranylgeranylacetone, inductive agent of chaperon molecule, was made to act on HGK, expression of mRNA of COX-2 and IL-8 was induced. In HGK, even 37℃ and heat treatment of 3 hours expression of mRNA of COX-2 was increased. In the heat treatment to HGF, expression of Hsp104 was not detected at 37℃ or 42℃. On the other hand, expression of Hsp90 increased at 42℃. Expression of Hsp70 and Hsp40 also increased at 42℃ heat treatment similarly. It is necessary to catch the influence of heat treatment in periosteum connective tissue in vivo based on the effect of warm temperature to gingival tissue.
