Research Abstract

本研究では、出芽酵母のRECQ遺伝子SGS1と遺伝学的に関連がある遺伝子を網羅的に集め、sgs1破壊株の表現型と対比させながら解析した。その中で特にMMS4遺伝子については、Mms4はRAD52組換え修復経路で働くが、Sgs1とは異なり、DNA傷害時の相同染色体間での組換えの上昇に関与しないことを明らかにした。また組換え中間体"HJ"を切断できる大腸菌RusAタンパク質をmms4変異株に発現させると。mms4株の薬剤感受性が相補されることを見いだした(Odagiri et al.,DNA repair,2003)。さらにMms4と複合体を形成するMus81のの欠損株mus81ではDNA傷害時に上昇する姉妹染色分体間の組換えが起こらないことをはじめて見いだし(未発表)、Mms4/Mus81の機能を知る上で、Keyとなる発見をしたと評価できる。
一方、脊椎動物細胞に存在する5つのRecQタンパク質(RECQL1,BLM,WRN,RTS,RECQL5)のうち、今まで機能未知のRECQL1,RECQL5がBLMの機能をバックアップすることをはじめて見いだした(Wang et al., Mol.Cell.Biol., in press)。さらに酵母でSgs1といっしょに機能するTop3に関して、ニワトリDT40 TOP3α単独株,TOP3α-BLM二重変異株を作製し解析した。その結果、BLM欠損株で観察される高頻度の"姉妹染色分体間の交換(SCE)"がDT40 TOP3α単独株でも見いだされ、SCEの抑制においてTop3αがBLMのco-factorとして働くことを明らかにした(投稿準備中)。DT40 TOP3α欠損株ではG2の初期にその細胞周期を停止し、細胞核は巨大化し、ヘテロクロマチン領域が弛緩しているデーターが得られ、Top3αがクロマチン高次構造の構築に働くことをはじめて示唆した(未発表)。
