Research Abstract

葉細胞の極性伸長制御系:横幅を制御するAN遺伝子をcloningの結果、CtBPのホモログをコードすることが判明した(Kim et al., 2002)。モチーフの機能についての作業仮説を確認中である。
一方、縦長を制御するROT3については、配列上最も類似したCYP90D1と共に、発現解析と光環境に対する応答性とについて解析した結果rot3-1変異体は、あらゆる光に対する葉身・葉柄伸長反応が失われていることが判明した。また暗所での葉柄の伸長パターンと、CYP90D1プロモーターの発現パターンとの間に、一定の連関が認められた。さらに、光による葉の展開過程の制御を詳細に解析し、Phytochromeを介した葉の展開制御には軸極性が認められること、細胞伸長と細胞分裂とは、それぞれ異なる制御をうけていること、葉柄と葉身とでは、各種光に対する反応が異なること、等を明らかとした(Tsukaya et al. 2002)。
葉身における細胞分裂の極性制御:ブラシノステロイドが細胞伸長のみならず、細胞分裂をも促進することが判明した(Nakaya et al. 2002)。さらに葉の横幅方向への細胞数が減少した細葉の変異体、an3について、原因遺伝子のクローニングを行なった。細胞分裂の極性制御遺伝子として、現在、機能を確認中である。
葉の左右対称性の制御系との関連:AS2遺伝子(Iwakawa et al. 2002)の変異の表現型を劇的に協調する新たな変異体bopについて、名大・町田研究室、韓国・南研究室と共同で解析した結果、BOPは、葉原基でのKNOX遺伝子発現抑制を司る新たな因子であることが判明した(Ha et al. 2003)。
野生植物での葉の極性軸制御:野生の種子植物で、個体間で葉の横幅に多型を示し、かつ葉の細胞数のみならず細胞サイズにも変異が認められる例を見いだし、初の例として報告した(Tsukaya 2002)。
以上、葉の形態形成メカニズムの解析を進めてきた成果をもとに、本年度、葉形態形成についての英文総説を複数発表した(Tsukaya, 2002a ; 2002b ; 2003)
